Background
Hepatocyte growth factor (HGF), also known as scatter factor (SF), is a multifunctional cytokine that regulates proliferation, survival, motility, angiogenesis, and tissue regeneration. HGF is synthesized as a single inactive precursor and proteolytically cleaved into an active heterodimer composed of α and β chains linked by a disulfide bond. Its biological activity is mediated through binding to the receptor tyrosine kinase c-Met, activating PI3K/AKT, ERK, FAK, and other signaling pathways.
The NK1 isoform is a naturally occurring splice variant comprising the N-terminal hairpin loop and the first kringle domain of HGF. This fragment retains receptor-binding capacity and exhibits partial agonist activity toward c-Met.
HGF and its NK1 variant are widely used in cell culture applications involving hepatocyte survival, endothelial biology, wound healing, mesenchymal cell differentiation, and regenerative medicine models. Aberrant HGF/c-Met signaling is implicated in tumor progression and metastasis.
