Introduction
A serum-free system for generating human intestinal organoids from pluripotent stem cells using a simple three-stage protocol through definitive endoderm, mid-hindgut spheroid, and intestinal organoid stages. The resulting organoids can be maintained long-term or cryopreserved, exhibiting cellular composition and organization representative of the developing intestinal epithelium and mesenchyme. Based on published formulations and optimized for improved efficiency and reproducibility across cell lines, this kit is suitable for studying intestinal development, cell biology, inflammation, regeneration, microbial interactions, disease modeling, and drug screening. Compatible with standard pluripotent stem cell maintenance media, with growth medium available separately for extended culture.
