Introduction
A complete serum-free and conditioned medium-free formulation for establishing and maintaining human intestinal organoids from crypt tissue. Isolated intestinal crypts rapidly form complex organoids featuring functional lumens enclosed by polarized epithelium containing all known adult intestinal epithelial cell types, with maturing organoids developing protruding buds resembling in vivo intestinal crypts. Supporting proliferation of epithelial stem cells from patient biopsies and expansion from fresh tissue or frozen stocks, applications include intestinal epithelium development and function studies, disease modeling, targeted screening, stem cell investigation, and regenerative therapy. Compatible with further differentiation using specialized differentiation medium.
