Introduction
A specialized medium supporting further differentiation of intestinal organoids in 3D culture or as 2D monolayers and air-liquid interface cultures. Starting cultures can be derived from intestinal crypts or passaged organoids maintained in growth medium, resulting in cultures containing physiologically relevant proportions of differentiated and stem cell populations that recapitulate crypt-villus axis diversity. Compared to conventional cell lines, differentiated monolayers exhibit greater barrier integrity, higher differentiation marker expression, and more representative morphology. Applications include intestinal epithelium development and function studies, disease modeling, compound screening, and regenerative therapy, with particular suitability for permeability assays and infectious disease studies due to apical surface accessibility. Requires growth medium for organoid initiation and expansion prior to differentiation.
Application
Cell Culture, Differentiation, Expansion, Maintenance, Organoid Culture
Feature
Enables versatile culture formats including 2D monolayers, facilitating direct access to the apical surface for straightforward host-pathogen interaction studies.
