Introduction

A system for generating choroid plexus organoids from human pluripotent stem cells for neural biomarker discovery and central nervous system permeability studies. These organoids model the blood-cerebrospinal fluid barrier with three-dimensional tissue structure and functional epithelium, providing controlled access for drug distribution studies and cerebrospinal fluid production research. Based on award-winning published protocols recognized for advances in animal reduction and replacement, the five-stage protocol includes embryoid body formation, neuroepithelial expansion, and patterning to choroid plexus-like epithelium. Mature organoids feature cystic structures filled with cerebrospinal fluid-like fluid surrounded by epithelium expressing choroid plexus-specific markers. Maturation components are available separately for extended culture beyond 40 days.
