Introduction
A complete, serum-free system for generating physiologically relevant blood vessel organoids with high efficiency and yield from human pluripotent stem cells. The streamlined protocol eliminates the need for serum screening and optimization, seamlessly integrating with standard pluripotent stem cell maintenance media. The resulting organoids develop extensive vascular networks featuring robust endothelial cell-pericyte interactions, characterized by CD31+, CD34+, CD144+, and KDR+ endothelial cells alongside PDGFR-β+, CD146+, SMA+, and NG-2+ pericytes. Designed for scalability, the kit supports high-throughput screening in 96-well formats and is suitable for tissue repair, regeneration studies, and vascular disease modeling. Maturation medium is available as part of the kit or separately.
