Introduction
A system for generating apical-out airway organoids in 15 days from human bronchial or airway epithelial cells using a simple scalable matrix-free protocol. These organoids provide access to the apical surface for infectious disease modeling and high-throughput drug screening in a three-dimensional in vitro system, producing polarized organoids with outward-facing differentiated ciliated cells in specialized microwell plates without cell culture inserts. The serum-free and bovine pituitary extract-free formulation maximizes experimental reproducibility. For optimal results, expand cells in compatible growth medium prior to differentiation.
