Introduction
A serum-free and bovine pituitary extract-free medium system for establishing and differentiating airway organoids from human bronchial epithelial cells. Following two-dimensional expansion in compatible growth medium, seeding medium initiates three-dimensional culture and differentiation medium generates morphologically relevant organoids. Fully differentiated organoids exhibit centralized lumens surrounded by polarized airway epithelium containing ciliated and goblet cells. Applications include lung disease modeling, drug efficacy and toxicity screening, and airway epithelium function studies.
